Dom was born in Willesden in 2000, beginning his football career with Arsenal as a central midfielder.

He originally signed schoolboy forms in 2012, The Gunners choosing to develop his talents at fullback before extending him a professional contract in 2018.

During the 2018/19 season, Thompson was a part of Arsenal's EFL Trophy squad on four occasions. He was also an unused reserve during their UEFA Europa League match against Ukrainian side FC Vorskla Poltava.

He joined Brentford on 7 August 2019, after nearly a decade in North London, getting his Bees debut six days later in a Carabao Cup defeat to Cambridge United at Griffin Park.

He first played in the Sky Bet Championship during Brentford's 3-0 win over Wigan Athletic in November 2019, coming on as a late substitute for Rico Henry. His FA Cup debut then came in January 2020, a 1-0 win over Stoke City, and he went on to play in the Fourth Round 0-1 defeat to Premier League Leicester City at Griffin Park.

On 4 January 2021, Thompson joined League One club Swindon Town on loan until the end of the 2020–21 season. Five days later, he made his debut with a start in a league match versus Ipswich Town and assisted each of Swindon's goals in the 3–2 win. He made 25 appearances during the remainder of a season which culminated in relegation to League Two.
